I'm aware of the thread that states there is no tutorial for creating flash profiles. That's not a problem for me . . . I'm quite familiar with flash. However, I have never designed a flash file specifically for MySpace.

My question comes in two parts. First, I have read that flash buttons do not work in MySpace and thus a transparent .gif overlay system is needed to make it all work. Is this truly the case?

Second, I've noticed a lot of layouts with standard content (top friends, comments, profile image) placed in unique places. Is this done in flash, or is the content placed using html code within MySpace?

To answer the first question, that isn't true. Or at least when I made flash layouts it wasn't.

And the second question, I don't know the answer, but I would suggest dissecting a lovemyflash layout. That should give you the answers you need.
